The 3.5L 2GR-FE V6 of late model Toyota and Lexus cars and SUVs can be tricky to bleed of air after coolant replacement. Toyota installed a bleed screw on this engine specifically to help with this. Located towards the front of the engine is a small 10mm brass fitting:
